Background: Porphyromonas gingivalis, a black-pigmented Gram-negative anaerobe, is widely implicated as an important
etiological agent in the pathogenesis of periodontal disease. Identification of P. gingivalis clinical isolate will advantage in research
efforts aimed at developing oral intervention methods against this bacterial species, and thus for prevention and treatment of
periodontal disease. Purpose: This study was to evaluate the ability of API 20A to identify P. gingivalis clinical isolate. Methods:
The methods were anaerobic culture and identification of P. gingivalis from bacteria Black-pigmented Gram-negative anaerobes
obtained from subgingival plaque of patients with chronic periodontitis, which based on API 20A system, Gram staining, colony
and microscopic morphology. Results: API 20A system detected P. gingivalis (P. asaccharolytica) clinical isolate obtained from
subgingival plaque with probing depth
≥ 3 mm. Colony and microscopic morphology showed that these bacteria were Gramnegative
strict anaerobic
coco-bacilli dark brown-black
pigmented. They grew
in culture
media forming convex, smooth glossy
colonies of 1-2 mm diameter. There were not differences when P. gingivalis clinical isolate were compared with P. gingivalis
ATCC 33277. Conclusion: The API 20A appears to be a promising method for identification of P. gingivalis clinical isolate.
