Karakterisasi Tingkat Embryogenesis dan Ekspresi Gen Gibberellin Family terhadap Perlakuan Chromium dengan Pendekatan Kultur In Vitro Tanaman Padi (Oryza sativa L.)
Abstract
Chromium is one of the dangerous heavy metals found in nature, manufacturing
industries and agricultural materials that has become a global concern and
recognized by the US EPA. Chromium has an impact on the environment, especially
plants because it can reduce crop yields due to inhibition of growth and seed
germination. As is known that seed germination is closely related to the GA
(Gibberellin) family which regulates seed germination. Several Gibberellin family
genes namely OsCPSI, OsKAO, OsGA2ox3, OsAmy1A, Os Actin were used to
detect the link between chromium and gibberellin family. This study attempts to
determine the screening of rice varieties that are resistant to abiotic stress of heavy
metal chromium using in vitro callus culture method. According to the findings,
Chromium treatment has an impact on plants seen from the parameters of
germination in vivo, browning on in vitro, stage of regeneration, cytochemical
multiple staining, and gene expression. Varieties that are resistant to chromium
stress are Getik and Sampit varieties.
