DINAMIKA POPULASI Xanthomonas oryzae pv. oryzae YANG DIINVESTASI PADA MEDIA TANAM PADI DENGAN APLIKASI EKSTRAK TAPAK LIMAN DAN BAKTERIOFAG XooM01
Abstract
Rice is the plant most widely cultivated by farmers, because rice is the main staple food for Indonesian people. However, rice productivity in East Java Province decreased from 56.68 to 56.47 Ku/ha. The decline in rice production was caused by bacterial leaf blight disease caused by Xanthomonas oryzae pv. oryzae. The Xoo pathogen can reduce yields by 70-80%, especially for rice that is cultivated continuously without crop rotation. This can cause the host or inoculum source to accumulate in the soil, which Xoo is known to survive in soil, plant debris, seeds and weeds. Alternative control can be done using Elephantopus scaber extract, bacteriophages and a combination of both. To find out the results of control, molecular research was carried out on the dynamics of the Xoo population in soil using PCR techniques and continued with semi-quantitative analysis. The lowest Xoo population decline occurred in the E.scaber treatment at 4 DPA. However, in all treatments there was an increase in population at 6 DPA, except for the control. Bacteriophage treatment and the combination of E.scaber + bacteriophage did not show a significant effect on reducing the Xoo population up to 6 DPA
