Ornamental plants still have high demand in Indonesia, especially orchids. Several types of
orchids are widely developed in tropical countries, but in Indonesia, the demand for
Dendrobium orchids still dominates so the need for orchid seedlings is also high. Orchid
seeds can only be grown with plant tissue culture techniques. In this technique, and the
acclimatization stage is a critical phase in determining the success of orchid seedling
growth. The environmental transfer from culture to the outside environment makes plantlets
adapt again and sometimes experience excessive transpiration and can interfere with
nutrient absorption. Chitosan is a simple derivative of chitin which is a natural organic
material and, a natural organic material that can function as a biofertilizer and bioimmuner.
The purpose of this study was to determine the response of orchid seedling growth to the
application of chitosan at the acclimatization stage and to see its success. This study used a
completely randomized design (CRD) with 1 factor the concentration of chitosan and 5
replications. Chitosan was given from a concentration range of 0 ppm, 2 ppm, 2.5 ppm, 3
ppm, 3.5 ppm, 4 ppm, 4.5 ppm, and 5 ppm. The data obtained were then analyzed for
variance with the F test at the 5% level. The treatment with significant effect was further
tested with Duncan Multiple Range Test (DMRT) at a 95% confidence level. The results of
this study showed that giving chitosan a concentration of 3 ppm at the acclimatization stage
had a significant effect on increasing plant height parameters, plant growth percentage, leaf
width, number of leaves, leaf length, root length, number of roots, and fresh weight of
Dendrobium sonia orchids.
