dc.description.abstract | Tissue culture plant propagation techniques are very promising, especially
for breeding in the business world produce seeds with uniform quality in a short
time. One type of plants can be propagated in vitro propagation of orchids are
Phalaenopsis sp L. To date more attractive than orchids other ornamental plants.
In tissue culture techniques can enhance the formation of chitosan Protocorn Like
Body (PLB) from vegetative explants orchid Phalaenopsis sp L. grown on solid
media. In addition the use of methods of propagation through the formation of
PLB is expected to address the problems of production of orchids in Indonesia,
the seed produced is not uniform, does not depend on time and quality of seeds in
accordance with the parent having superior properties. Sterilization of tools and
materials are things that must be considered in the current study, sterilization can
be done by means of a temperature mengautoclave 1210 C submarine 2 hours at a
pressure of 17.5 psi. The research method using the Completely Randomized
Design (CRD), with chitosan concentration of 0, 5, 10, 15, 20 and 25 ppm with
each treatment as much as 3 replicates. From the research results can be seen that,
with the provision of 15 ppm chitosan in weight parameters, the number and
diameter of PLB showed better results, whereas at a concentration of 5 ppm on the
parameters of the number of plantlets, leaf number, root number and root length
showed better results than on the other treatment. | en_US |
