Abstract
SARS-CoV-2 (Severe Acute Respiratory Syndrome Coronavirus 2) is the virus that
causes COVID-19 (Coronavirus Disease 2019). The virus was first identified in
late 2019 in Wuhan, China, and has since spread around the world, causing a
global pandemic. One of the efforts to prevent the expansion of the spread with
the development of targeted vaccine manufacturing, COVID-19 vaccines in
general are spike proteins. important research in the development of COVID-19
vaccines successfully created antibodies that can target SARS-CoV-2 spike
proteins and show potential as vaccine candidates. This study provides very
important insights in the development of a COVID-19 vaccine, one of which has
been carried out in the form of research in the production of recombinant proteins
encoding the SARS-CoV-2 spike protein of the Indonesian strain. Recombinant
protein production is carried out in silico through a bioinformatics approach. The
protein was also produced in vitro through GeneticaScience. The effectiveness of
the recombinant protein as an antigen has been studied in vivo to determine its
effectiveness in inducing immunity and then tested on the liver organ of White
Rats (Rattus norvegicus) test animals and then analyzed the concentration levels
using the Bradford method. Protein profiling is done by extracting proteins from
the organ then tested using the SDS-PAGE method. The results of SDS-PAGE
were then analyzed using ImageJ software and then tested using the SDS-PAGE
method.
