Abstract

This study aims to determine the effect of cocoa leaf extract (Theobroma cacao) on the histological picture of the seminiferous tubules of male mice (Mus musculus). The background of the study was based on the need for alternative male contraception using natural ingredients, because the current choice of male contraception is still limited. Cocoa leaves were chosen because they contain active compounds such as flavonoids, alkaloids, tannins, saponins, and theobromine which have antifertility potential. The study used 24 male mice with 4 treatment groups, namely one control group using distilled water, P1 extract dose of 200 mg/kgBW, P2 extract dose of 300 mg/kgBW, and P3 extract dose of 400 mg/kgBW. The extract was given orally for 14 days. The parameters observed included testicular weight, seminiferous tubule diameter, and the number of spermatogenic cells (spermatogonium cells, primary spermatocyte cells, and spermatids). Data analysis was carried out using the One Way Anova Test with a confidence level of 95%. The results of the study showed that the administration of cocoa leaf extract (Theobroma cacao) did not significantly affect all observed parameters (P>0.05). The final product in the form of a book chapter has been validated and declared suitable as a scientific information media that can be accessed by the general public.
