The purpose of this study was to obtain the optimal composition of hydroxypropyl methylcellulose (HPMC) K4M (A) and sodium alginate (B) in gastroretentive behaviour of theophylline tablets. The tablets were formulated as floating mucoadhesive system and prepared by wet granulation. Factorial design is an optimization method used in this study. Four formulas were obtained that are: F1 (10% A &10% B ), F2 (20% A & 10% B), F3 (10% A & 30% B), and F4 (20% A & 30% B). The respons of this study are behaviour of floating, mucoadhesive, and swelling. HPMC was very dominant factor in shortening lag time and in increasing tablet’s size. Sodium alginate was very dominant factor in improving mucoadhesive behaviour. Both HPMC and sodium alginate were dominant in improving tablet’s floating duration time. The optimized formulation are 10-20% of HPMC and 24,013-30% sodium alginate. The optimized formula followed zero order kinetics in it’s drug release, DE value was 54,023% and 16,967 hours was needed to release 100% theophylline from tablets.
