Potensi Protease Ekstraseluler Fictibacillus barbaricus GaN 2 Asal Pantai Papuma Jember sebagai Agen Fibrinolitik

Abstract

Marine bacteria are a promising source of bioactive compounds and extracellular enzymes due to their adaptation to unique marine environments. Among these enzymes, fibrinolytic enzymes have attracted considerable attention for their ability to degrade fibrin, making them potential candidates for the development of alternative thrombolytic agents in the treatment of thrombosis. This study aimed to determine the fibrinolytic activities of the crude protein extract of Fictibacillus barbaricus GaN 2 isolated from the coastal waters of Papuma Beach, Jember. Prior to fibrinolytic evaluation, the identity of the isolate and its proteolytic activity were confirmed through morphological characterization, Gram staining, and proteolytic assays on Skim Milk Agar (SMA). Extracellular proteins were extracted by ethanol precipitation, quantified using the Bradford method, and evaluated for fibrinolytic activity using the fibrin plate assay with nattokinase as a positive control and 1% DMSO as a negative control. The isolate was confirmed to exhibit morphological characteristics and proteolytic activity consistent with previous reports. The crude protein extract contained a protein concentration of 151.925 ng/µL and exhibited positive fibrinolytic activity with a fibrinolytic index of 1.22 ± 0.19 after 9 h of incubation, corresponding to approximately 56.2% of the activity of the positive control, nattokinase (2.17 ± 0.01). These findings demonstrate that the crude protein extract of F. barbaricus GaN 2 possesses fibrinolytic activity, indicating its potential as a source of fibrinolytic enzymes
