Abstract
Tobacco (Nicotiana tabacum L.) is a plantation commodity that plays a vital role in Indonesia's agricultural sector. Tissue culture is crucial for developing superior seeds with optimal quality outside the harvest season. Furthermore, the goal is to produce seeds that are identical to the parent plant and free from disease. The success of tissue culture can be supported by the application of plant growth regulators such as Naphthalene Acetic Acid (NAA) and Furfuryl Amino Purine (Kinetin). The second treatment involves the placement of tobacco leaf explants. This study aimed to determine the optimal concentration of the combination of NAA and kinetin, as well as the placement of the planting position, for the regeneration of tobacco leaf explants. This research was conducted from January to May in the Ecophysiology and Tissue Culture Laboratory, Agronomy Study Program, Faculty of Agriculture, University of Jember. The experimental design used was a Completely Randomized Design (CRD) with two factors : (0.25; 0.5) ppm NAA and (1.5; 2.5; 3.5) ppm kinetin and the placement of the leaf explant planting position, namely the abaxial side up and the adaxial side up. The interaction of the combination of NAA and kinetin and the placement of the planting position affected several parameters. However, the administration of a concentration of 0.5 ppm NAA and 1.5 ppm kinetin (H7) with adaxial placement facing upwards (P2) gave the best results in plant regeneration.
