PENGEMBANGAN DAN VALIDASI METODE KLT DENSITOMETRI UNTUK PENETAPAN KADAR INULIN DALAM EKSTRAK AIR UMBI BENGKUANG

A simple, sensitive, selective and specific, precise and accurate TLC
densitometry method has been developed and validated for determination of inulin in
water extracts of yam bean
as the stationary phase and using
glacial acetic acid :methanol p.a: aquabidest

10 min, then stained with a mixture of aniline: diphenylamine: phosphoric acid
C for 15
minutes. Quantitative evaluation was performed by measuring the absorbance of the
analyte spot at 380 nm. The linear regression data for the calibration plots of inulin
showed good linear relationship with r = 0,99610 in the concentration range 6002400
ppm.
The
minimum detectable amounts were found to be 35,515 ppm and the
limits of quantitation were found to be 118,383 ppm. Repeatability precision, as RSD
was 2,754% and Intermediet precision, as RSD was 1,935%. Accuracy of the method
was determined through recovery studies by adding known quantities of standard
drug to the pre analyzed test solution and was found to be 100,161% ± 1,839%. This
TLC Densitometry method is selective and specific, precise, and accurate and can be
used for routine analysis of inulin.
