The glass ionomer cemebt has been used as fissure sealant, because it contains fluoride. Fluor application could reduce caries processes by preventing enamel decalcification. The aim of the study was to know tooth enamel decalcificatiion degree after glass ionomer application on pit and fissure of tooth surface in-vitro. The samples of this study were maxillary first premolars post-orthodontic extraction, and were divided into two groups. The first group was applied with type III glass ionomer cement on pit and fissure of tooth surface. The second group was applied without type III glass ionomer cement. Enamel decalcification was examined by spectrophotometer. Data was analysed using t-test. Te result of the study revealed that there was a significant difference (p<0,05) between the first and the second group, in which calcium dissolved on the tooth that was applied with glass ionomer cement lower than the tooth without glass ionomer cement application
