    Abstract
    The new paradigm that has been developed for the treatment of periodontal infection is host modulation therapy, therapy aimed at improving the immune response. Infection therapy which has been used only aim to kill the bacteria without regard to the balance of the host response. Therefore, it is necessary to develop a drug therapy to improve the resilience of neutrophils in fighting bacterial infections . Purpose . This study is aimed to potential of Robusta coffee bean extract in modulating the adhesion of P. gingivalis on neutrophils as well as its effect on the viability of neutrophils exposed to P. gingivalis . Method . Experimental studies in vitro using isolated neutrophils samples from human peripheral venous blood . 1) Test the adhesion of P. gingivalis in neutrophils. Neutrophil suspension coated on plastic micro plates ( 24 wells ) were given a cover slip , and incubated robusta coffee bean extract concentration of 100 %, 50 %, 25 %, as control neutrophils incubated RPMI . Furthermore, neutrophils exposed to P. gingivalis 3x108 CFU / ml . Fixation with methanol and Giemsa dye staining and counting the adhesion of P. gingivalis in neutrophils microscopically. 2) Test the viability, in brief neutrophil suspensions were divided into four test groups ie , incubated robusta coffee bean extract concentration of 100 %, 50 %, 25 % and as control neutrophils incubated RPMI . Incubation was performed for 18 hours in a roller tool . Furthermore, neutrophils exposed to a suspension of P. gingivalis 3x108 CFU / ml and incubation was 1 hour, then test the viability of using trypane blue dye. Neutrophils were viable (living) does not absorb trypane blue so it looks clear or transparant, while the dead neutrophils trypane absorbs blue color. Results a). Neutrophil adhesion smallest in group IV (coffee extract 100 %) , b). Viability of neutrophils are most often found in group III (50 % coffee extract). Conclusion. Robusta coffee bean extract can reducing the amount of adhesion P . gingivalis on neutrophils as well as increasing the number of neutrophils in cell viability.
