Abstract The aim of this research was prepared a captopril buccal mucoadhesive drug delivery system. Buccal mucoadhesive tablet of captopril were developed to prolong release time and increase its bioavailability. The tablet were prepared by direct compression technique, using polimer HPMC and chitosan as buccal mucoadhesive system. The effect of HPMC and chitosan on drug release profile and mucoadhesive properties were investigated. Factorial design method was employed to create the optimum composition of HPMC and chitosan and used to determinate the effect of HPMC and chitosan and their interaction on mucoadhesive and dissolution efficiency. The optimum formula which obtain several criteria was investigated with overlay plot. The linier regression analisys and model fitting showed that all this formulation followed Higuchi model, which had highest value of correlation coefficient (r). Optimum compotition based on overlay plot for each tablet is 302,6 mg - 330 mg of chitosan at 90 mg of HPMC and 77,65 mg - 90 mg of HPMC at 330 mg chitosan.en_US
