Abstract

Farmers have long utilized synthetic chemical pesticides to control Spodoptera. litura. However, these chemical insecticides can lead to various negative impacts. A safer alternative is the Spodoptera litura Nuclear Polyhedrosis Virus (SlNPV). The efficiency of SlNPV biopesticide production in vivo can be enhanced by optimizing the inoculum concentration dose and the larval stadia phase at which the virus is inoculated. This study aims to analyze the interaction effect of virus concentration and larval instar stage of S. litura on mortality and the production of Polyhedral Occlusion Bodies (POBs). The method employed was a factorial completely randomized design (CRD) with two treatment factors. The first factor was SlNPV concentration (105, 106, 107, 108), and the second was the larval instar stage (Instar 3, 4, 5). Each treatment combination was repeated three times, resulting in 36 experimental units. ANOVA results indicated that the interaction between SlNPV concentration and larval instar stage did not significantly affect S. litura mortality (Sig = 0.527), but had a significant effect on POBs production (Sig = 0.000). Based on the results of this study, it is recommended to use an SlNPV concentration of 105 on larval instar stage 3 is optimal and efficient for S. litura mortality. The use of SlNPV at C4 (108) concentration in instar 4 is recommended to produce the highest POBs/ml.
