Abstract

Background: Traumatic ulcers have a prevalence of 6.5% at the Oral Medicine Clinic of the University of Jember Dental and Oral Hospital. Bacterial adhesion and colonization on oral lesions can delay the healing process, highlighting the need for effective treatments to control bacterial growth and accelerate wound healing. Tobacco leaves, which are abundantly available in Jember, contain active antibacterial compounds such as alkaloids, saponins, flavonoids, and essential oils, making them a potential natural therapeutic agent for the management of traumatic oral ulcers. Objective: To determine the effectiveness of tobacco leaf extract gel in reducing bacterial colonies in traumatic ulcers of Wistar rats. Methods: This study is an experimental research using a repeated post-test measurements design, consisting of four groups ( 2% CMC-Na gel; 5% tobacco leaf extract gel;10% tobacco leaf extract gel; 20% tobacco leaf extract gel). The observed parameter was the number of bacterial colonies, which was assessed by swab sampling of traumatic ulcers in Wistar rats on days 1, 3, 7, and 14. Results: Based on the Mann–Whitney test, there was a significant difference (p ≤ 0.05) between the 10% and 20% concentrations of tobacco leaf extract gel in reducing bacterial colony counts in traumatic ulcers of Wistar rats. Conclusion: Tobacco leaf extract gel is effective in reducing bacterial colony counts in traumatic ulcers of Wistar rats at concentrations of 10% and 20%.
