To analize the effects of caffeine on improvement osteoclastogenesis and orthodontic tooth movement. Material and methods: The research method was experimental laboratories, 16 rats were divided into 2 groups. Group C: the rats were applied with Orthodontic Mechanical Force (OMF) and 2 ml of distilled water, group T: OMF and caffeine of 1,37mg/100g BW in 2 ml of distilled water. OMF was conducted by applying ligature wires in diameter of 0.20 mm on the molar-1 (M-1) of right maxilla and both incisivus of maxilla. Subsequently, the M-1 of right maxilla was moved to mesial using Tension Gauce to generate power of 10g/cm2 with Ni-Ti orthodontic closed coil spring. Histological examination was conducted to calculate the number of osteoclasts. Increased tooth movement in the rats was measured by measuring from the tip of the distal of M-2 to the end of M-1 mesial of right maxilla lessened by mesiodistal width of right upper M-1 and M-2 using digital caliper. Results: the administration of caffeine steeping proved to effectively increased the number of osteoclast in both compression area and tension area more than those which were not administered with caffeine (p <0.05). The number of osteoclast in compression area more than the tension area (p <0.05).The administration of caffeine improved the movement of M1 of right maxilla compared to those not administered with caffeine (p <0.05). Conclusion: the administration of caffeine increased osteoclastogenesis and improved the movement of the teeth, therefore it may be an alternative to accelerate orthodontic treatmenten_US
