Background: Periodontitis is an inflammatory disease affecting tooth supporting
tissues. This disease is caused by Porphyromonasgingivalis and can extend into deeper
tissues involving alveolar bone. Inflammation is a host defense response that can lead to
gingival epithelium destruction. Robusta coffee seed contain caffeine and chologenic
acid which can act as anti-inflammation and antioxidant agents. Purpose: The aim of this
research was to analyze the effect of robusta coffee seed extract gel administration on
the density of gingival epithelium in P.gingivalis induced periodontitis rat models. Methods:
Thirty-two samples of rats were divided into 8 groups; negative control group (normal),
positive control group (periodontitis + placebo gel), 25% treatment group (periodontitis +
25% robusta coffee seed extract gel), 50% treatment group (periodontitis + 50% robusta
coffee seed extract gel) in which each of them was further divided into day 1 euthanasia
group and day 7 euthanasia group. Each group consisted of 4 samples. After that HE
staining was carried out for epithelium density was measured using software imageraster.
Results: K4 (50% robusta coffee seed extract gel group) has higher mean of epithelium
density than those of group 2 and 3. Conclusion: The administration of robusta coffee seed
extract gel increased the gingival epithelium density of periodontitis rat model.
