    Abstract
    Abstract The aim of this research was to develop an optimized gastric floating drug delivery system (GFDDS) of theophylline. Floating tablet of theophylline were developed to prolong gastric residence time and increase its biovalability. The tablet were prepared by direct compression technique, using polimer HPMC and carbopol as floating system. The produced tablets were tested for physical properties, such as uniformity of weight, tablet hardness, floating test and in vitro dissolution test. The effect of HPMC and carbopol on drug release profile and floating properties were investigated. The results indicate that HPMC and carbopol affected floating lag time, floating duration time and released properties of theophylline. Addition concentration of HPMC increase floating lag time, but addition of carbopol decrease floating lag time. The effect of polymer toward floating duration time is unknown because the results is uniform. Dissolution efficiency is decrease by addition of polymers. Factorial design method was employed to create the optimum composition of HPMC and carbopol and used to determinate the effect of HPMC and carbopol and their interaction on floating properties and dissolution efficiency. The optimum formula which obtain several criteria was investigated with overlay plot. The linier regression analysys and model fitting showed that formula 2 dan 4 followed zero order, and formula 1 and 3 followed Higuchi model which had highest value of correlation coefficient (r). Optimum compotition based on overlay plot for each tablet is 119,06 mg – 120,00 mg of HPMC at 20 - 30 mg of carbopol.
