Abstract: Klebsiella pneumoniae is a bacteria of the genus Klebsiella in the family Enterobactericeae. Klebsiella pneumoniae causes serious infectionst. Virulence factors are important in the initial attachment of bacteria in infecting hosts are pili and Outer Membrane Protein (OMP). Pili as a virulence factor can be strongly attached specifically and able to avoid cleaning done by host cells. The adhesion ability of bacterium is influenced by its ability to agglutinate erythrocytes. Based on the preliminary studies that have been conducted, molecular weight of the pili protein obtained from SDS-PAGE electrophoresis is 96.4 kDa has a thick and high concentration band, no studies have shown that the pili protein 96.4 kDa is a hemagglutinin and adhesin protein. The purpose of this study is to determine the role of 96,4 kDa protein pili of Klebsiella pneumoniae as hemagglutinin and adhesin protein. The results of hemagglutination test using mice erythrocyte cells obtained the highest hemagglutination titer for the molecular weight of pili protein 96.4 kDa is in titer 1. Adhesion index with dilution titer had a significant relations which was 0.016 (p <0.05) with r = - 0.847. It was concluded that 96,4 kDa pili protein of K.penumonia is a hemaglutinin and adhesin protein.
