dc.description.abstractThe development tomato (Lycopersicon esculentum) high sucrose production can be done by genetic engineering methods through transformation techniques. Tomato transformation and regeneration were analysed and optimized. Cotyledon and axillary bud explants from Lycopersicon esculentum cv. Zamrud and Liontin, were infected by Agrobacterium tumefaciens strain LBA4404 harbouring the neomycin phosphotransferase (NPTII) reporter gene, CaMV35S promoter and SoSPS1 gene. Explant were used as target tissue age of 14, 15, 16 days. Selection of transformed explants was performed on MS medium containing 0, 25, 50, 75 and 100 ppm of kanamycin. The aim study is to determine the efficiency of transformation and the increase in sucrose content in transgenic tomato plants transformed with SoSPS1 genes using Agrobacterium tumefaciens. The results showed that the Zamrud variety is better than Liontin. Cotyledon explants showed better results than axillary buds. Kanamycin concentration of 50 ppm indicates the plant has been selected. Cotyledon explant age 14 days to improve transformation efficiency of 72%. Based on PCR results showed that the plasmid pKYS-SoPS1 already integrated in the explant. Transgenic tomato plants have been obtained as many as 8 transgenic plants.en_US
