Abstract: High psychological and physical distress work can stimulates cellular and tissue damage by corticosterone secretion, included in periodontal cells and tissues. If this conditions not receives suitable treatment, it alters tooth mobility or premature loss. For maintaining body homeostasis, the cells and tissues will responses with many mechanism to prevent cells and tissue damage, ie by HSP-60 and HSP-90. HSP-60 and HSP-90 have been shown to play pro- and anti-apoptotic roles, respectively. The objective of this research is to determine the role of HSP-60 and HSP-90 on the periodontal ligament stem cells/PDLSC (multipotent periodontal cells) related to distress work. The results of this research are expected to obtain information about (a) the role of corticosterone in production HSP-60 and HSP-90 protein, (b) the role of HSP-60 and HSP-90 to repair cell damage in chronic labor distress. The Conclusions are (1) increase corticosteroids (Elisa) in psychological and psychological+Physics distress work due to increasing production HSP-60 and HSP-90 on PDLSC (IHC), (2) increasing in the expression of HSP-60 resulting in a decrease in the number of fibroblasts (HE). Key Words: Distress, PDLSC, HSP-60, HSP-90, Fibroblast
