Abstract: Ferns are vascular plants that have neither seeds nor flowers and reproduce via spores. Ferns are often used in traditional medicine since they have various health benefits. Tectaria heracleifolia is one such fern. In traditional medicine, Tectaria heracleifolia is used for treating urinary-related diseases, diabetes, cough, headaches, vertigo, and more. Despite that, Tectaria heracleifolia has not been extensively studied. This study aims to identify the secondary metabolite groups, total phenolic content, total flavonoid content, and antioxidant activity of the methanol extract of Tectaria heracleifolia. Phytochemical screening was done with thin-layer chromatography (TLC) and it was found that the methanol extract of Tectaria heracleifolia does not contain alkaloids, triterpenoids, or saponins, but it does contain phenolic compounds and flavonoids. The total phenolic content of the methanol extract of Tectaria heracleifolia was determined with the Folin-Ciocalteu method and found to be 217.7075 ± 1.3482 mg Gallic Acid Equivalent/g extract. The total flavonoid content was determined by the AlCl₃ colorimetric method and was found to be 52.8549 ± 0.5943 mg Quercetin Equivalent/g extract. The antioxidant activity of the extract was determined with DPPH and found to have an IC50 value of 27.2110 ± 0.4609 µg/mL.
