Abstract: Indonesia is a megadiverse country with the second largest biodiversity in the world. As many as 31,750 plants and fungi grow in Indonesia, and 7,000 of them have medicinal properties. One type of biodiversity that has the potential to be used as medicine is lichen. Lichen is a mutualistic organism consisting of fungi (mycobionts) and algae (photobionts). Most types of lichen contain unique secondary metabolites in the form of depside, depson, depsidone, and dibenzofurane, which have important pharmacological activities. The purpose of this study was to determine the molecular structure of the methanol extract compound of the lichen Leptogium corticola (Taylor) Tuck from the isolation results. The initial stage of this study was extraction using the maceration method. Then, the fractionation process used the VLC (Vacuum Liquid Chromatography) method, fraction grouping was carried out using TLC (Thin Layer Chromatography) with n-hexane: ethyl (9:1) eluent as much as 4 mL. Fractions that were not pure were isolated using HPLC (High Performance Liquid Chromatography). Identification of compounds in pure isolates was carried out using Nuclear Magnetic Resonance Instrument (1D-NMR and 2D-NMR). The methanol extract obtained was 6,79 % and the HPLC isolate was in the form of black crystals as much as 4,38 %. The results of the NMR spectra identification analysis obtained the compound methyl 2-hydroxy-4,6-dimethylbenzoate. This study is the first study to successfully isolate the compound structure from the lichen Leptogium corticola (Taylor) Tuck.
