Abstract: | Quercetin is a flavonoid from Allium cepa (shallot) which has biological activity such as antiinflammation, antidiabetic, anticancer, and antioxidant. Therefore, there are a lot of research on quercetin as a potential compound that can be used to treat various diseases. This research requires quercetin in isolated form. Molecularly Imprinted Polymers (MIP) is an effective method for separating compound from methanol extract of shallot. In this research, MIP for quercetin was synthesized using a bulk polymerization method through a noncovalent approach. A macroporous polymer was synthesized using D-arabinitol as a template, acrylamide as a functional monomer, ethylene glycol dimethacrylate (EGDMA) as a cross-linker, methanol as a porogen, and benzoyl peroxide as an initiator. MIP was synthesized in several formulas with a different molar ratio of functional monomers and cross-linker. Synthesis process was carried out in a closed system at 60℃ for 24 hours. After the process of polymer synthesis, quercetin was removed by a mixture of methanol and acetic acid (4:1, v/v). NIP was synthesized using same method as MIP without the addition of quercetin as a template. Fourier-transform infrared spectroscopy (FT-IR) and scanning electron microscopy (SEM) were used to characterize the MIP produced. A batch rebinding assay was used to test the binding efficiency of each formula. Batch rebinding test result revealed that MIP 4 had the highest binding capacity at 0,64 mg/g. % recovery was used to determine the extraction efficiency of MIP against quercetin. % recovery of quercetin MIP was 86,81%. |
