Background: Tooth extraction is an action often carried out by dentists in the field of dentistry. Tooth extraction causes injuries and pain involving hard and soft tissue. Pain is always associated with the area where inflammation occurs. Pain makes a person take anti-inflammatory drugs so that the pain disappears. However, many herbal plants can be an alternative to chemical drugs which are believed to be used as anti-inflammatory drugs. One of these herbal plants is purple leaf (Graptophyllum pictum L. Griff) which can speed up the wound healing process. One of the cells that plays a role in wound healing in the inflammatory process is macrophages. Macrophages first appear at the time of injury, namely in the first 48-96 hours, and will reach a peak on the 5th day. This study aims to determine the potential of administering purple leaf extract in increasing macrophages after Wistar rat tooth extraction. Method: This research uses the Post Test Only Control Design method and an in vivo experimental research type. This research used male Wistar rats weighing 150-200 grams. The experimental animals were divided into three research groups: day 3, day 5, and day 7 groups. The data results were processed using the IBM SPSS Statistics 25 application. The data results were analyzed using the One Way Anova test, followed by the Post Hoc LSD test. Results: The test results showed that there were significant differences in each research group. Conclusion: Based on this research, there was potentially affecting the number of macrophages in the wound healing after tooth extraction in Wistar rats given purple leaf extract.en_US
