Background: Traumatic ulser are lesions of the oral cavity that are commonly encountered. Usually caused by trauma such as biting of the mucosal wall due to chewing and drinking and hot food. Long-term use of corticosteroid anti-inflammatory drugs causes immune system resistance and epithelial cell atrophy. Red pomegranate extract gel (Punica granatum Linn) contains flavonoids, saponins, tannins and ellagic acid which can increase fibroblast activity so as to accelerate ulcer healing. Purpose: The aim of the research was to determine the effect of red pomegranate (Punica granatum Linn) extract gel with a concentration of 75% on the increase in fibroblasts in healing traumatic ulcers of Wistar rats (Rattus norvegicus). Methods: This type of research was experimental laboratory with 24 Wistar rats divided into a control group on day 3, day 5 and a treatment group given red pomegranate extract gel concentration of 75% on day 3 and day 5 which were euthanized on 1 day after day 3 and day 5. Each research sample, a traumatic ulcer wound will be created on the lower left buccal mucosa using an amalgam stopper heated over a fire for 30 seconds and applied for 1 second without pressure. After the specified time, the Wistar rats are then euthanized and their tissue were processed and stained with Hematoxylin and Eosin, and then the fibroblast cell was counted using a binocular light microscope at 400x magnification. Data were analyzed using one way Anova and LSD (Least Signifience Different). Result: The result of the one way Anova test showed significant differences in the number of fibroblast in all sample groups (p<0.05) and the LSD test showed significant differences between the groups (p<0,05). Conclusion: It can be concluded that red pomegranate (Punica granatum Linn) extract gel with a concentration of 75% has an effect on increasing fibroblasts in the healing process of traumatic ulcers.en_US
